In this issue of Cell, Wan et al. (2019) track comprehensively the development of individual neurons, along with their activity, during zebrafish spinal cord development. They find that mostly motor neurons are the founders of initially small neuronal-activity ensembles, coalescing into larger populations establishing the first motor patterns.

Slow oscillations and delta waves are neuronal activity rhythms that hallmark sleep, but until now their respective functional roles have been impossible to tease apart. Utilizing a closed-loop optogenetic approach in rats, Kim et al. (2019) dissociated the functions of these two canonical rhythms, showing they support the consolidation and forgetting of memories, respectively.

Type 2-low asthma represents a critical unmet need and an opportunity for new drug discovery. Here, Maun et al. demonstrate that increased mast cell tryptase levels are associated with asthma severity independent of type 2 inflammation and that increased active tryptase alleles are associated with decreased clinical responses to anti-IgE treatment. They generated a noncompetitive inhibitory anti-tryptase antibody with in vivo activity in humanized mouse and cynomolgus monkey models as a clinical candidate for severe asthma treatment.

Wan et al. reconstruct neurogenesis and the emergence of coordinated neuronal activity at the single-cell level in the zebrafish spinal cord by tracking neuron lineages, movements, molecular identities, and activity in the entire developing circuit. They find that functional maturation of neurons is stereotyped, based on birth time and anatomical origin, and that early motoneuron activity leads ensembles that synchronize globally, based on network size.
